Introduction | Leucine-rich repeat kinase 2 (LRRK2), and PARK8 (derived from early recognition of Parkinson's disease), is a kinase gene encoded by human LRRK2. LRRK2 is a member of the leucine-rich repeat kinase family. Variations in this gene are associated with increased risk of Parkinson's disease and Crohn's disease. The expression of mutant LRRK2 induces apoptosis in neuroblastoma cells and mouse cortical neurons. The expression of LRRK2 mutants associated with autosomal dominant Parkinson's disease leads to shortening and simplification of dendritic trees in vivo and in cultured neurons. RRK2 is also a substrate for partner-mediated autophagy. |
